How do we study orphan GPCRs? Diagram

As part of our efforts to deorphanize orphan GPCRs, we have recently developed and optimized a novel cell-based assay called the Gz-Enhanced Signal Transduction assaY (GzESTY). This assay provides increased sensitivity for studying Gi/o/z-coupled receptors by redirecting their activation to an increase in intracellular cAMP levels. By measuring cAMP in real time, we can accurately quantify ligand-induced activation of GPCRs. GzESTY is currently being used to isolate both endogenous and synthetic ligands for orphan GPCRs. Additionally, in collaboration with the lab of Dr. John D. Lueck (University of Rochester), we have packaged GzESTY into an all-in-one plasmid to facilitate transfection protocols and improve the reproducibility of large-scale experiments.
